Understanding Semen Detection, DNA Extraction & Comparison, and Complete Infidelity Test Reports

Infidelity DNA testing is designed to analyze biological material that may be present on personal items.

Unlike paternity or sibling testing, which evaluates a biological relationship between individuals, infidelity testing focuses on detecting biological evidence and, when possible, comparing DNA profiles.

Below are examples of how results are structured for each type of infidelity DNA test.

Semen Detection Test Result Examples

A Semen Detection Test is a qualitative screening test.

This test looks specifically for the presence of semenogelin, a protein that is highly specific to human semen. Semenogelin detection is widely used in forensic analysis because it is highly specific and does not cross-react with other biological substances.

It is important to understand:

  • This test does not produce a DNA profile

  • This test does not identify a person

  • This test does not determine how much semen is present

  • This test only reports whether semenogelin was detected

The Two Possible Results

Positive Result

If semenogelin is detected, the report will state that the sample tested positive for the presence of semen, semen was detected on this item.

Negative Result

If semenogelin is not detected, the report will state that the sample tested negative for the presence of semen, semen was detected on this item.

DNA Extraction and Comparison Test Result Example

DNA Extraction and Comparison testing is designed to determine whether biological DNA can be recovered from a submitted item and, if so, whether that DNA is consistent with a provided reference sample.

It is important to note:

  • DNA recovery is not guaranteed

  • The test looks for and reports whether interpretable DNA is present

  • If DNA is detected, a genetic profile may be developed

  • If comparison samples are provided, the laboratory can evaluate whether profiles are consistent or not consistent

The results may report:

  • DNA detected

  • No reportable DNA detected

  • A DNA profile consistent with a provided reference sample

  • A DNA profile not consistent with a provided reference sample

  • Sex of the DNA profiles generated

  • Inconclusive findings if the DNA is insufficient or too degraded to use as a comparison

This test evaluates whether biological material may be present and whether it may be consistent with a specific individual when a reference sample is available.

Complete DNA Extraction and Comparison with Semen Detection Result Example

The Complete Infidelity DNA Test combines both analyses:

  • Semenogelin screening (qualitative semen detection)

  • DNA extraction and comparison analysis

This combined report provides:

  • A result indicating whether semenogelin was detected

  • An evaluation of whether interpretable DNA was recovered

  • A comparison analysis if reference samples were submitted

Because both tests are performed, the report may include:

  • Positive or Negative semen detection results

  • DNA detected or no interpretable DNA detected

  • Consistent or not consistent comparison conclusions

  • Sex of the DNA profiles generated

  • Inconclusive findings if the DNA is insufficient or too degraded to use as a comparison

This option provides the most comprehensive analysis available within infidelity DNA testing.

Important Considerations About Infidelity DNA Test Results

Infidelity DNA testing is forensic in nature. The ability to detect semenogelin or develop a DNA profile depends on many factors, including sample condition and biological material present.

Results are reported based on the scientific data obtained during analysis. Conclusions are carefully written to reflect what the evidence supports and do not make assumptions beyond the laboratory findings.

Why Our Infidelity DNA Test Results Are Reliable

At the core of everything we do is one simple principle: accurate answers matter.

Our laboratory partners:

  • Use validated forensic screening methods

  • Follow strict quality control procedures

  • Perform relationship testing twice when applicable

  • Maintain high-level laboratory accreditations

This ensures that all findings are reported accurately and responsibly.
